Cutting one amino acid extended male mouse lifespan by 23%

Lifelong valine restriction improved healthspan in both sexes but lengthened life only in males, in a Nature Aging study.

A study in Nature Aging reports that restricting dietary valine — one of the three branched-chain amino acids — across the whole life of C57BL/6J mice raised median male lifespan by 23%. It did not extend female lifespan at all. Both sexes showed reduced frailty, lower cancer prevalence and a smaller burden of senescent cells, along with improved leanness and glycemic control across ages.

The sex split is the interesting part rather than an inconvenience. The authors link the male-only lifespan effect to a male-specific downregulation of PI3K-Akt signalling, alongside a liver gene module enriched in mitochondrial pathways and higher mitochondrial respiration in valine-restricted males. That gives a mechanism to test rather than a correlation to admire, and it points at compounds that could mimic the restriction without requiring anyone to redesign their diet for life.

Why this belongs under HEALTH's preventive care and longevity heading: dietary amino acid restriction is unusually cheap to study and, if a mimetic works, unusually cheap to deliver. Most longevity interventions that reach humans are expensive biologics. This class is not.

The caveats are large and belong in the foreground. These are mice, on a lifelong intervention started early, in one strain. The healthspan benefits appeared in both sexes but the lifespan benefit did not, and a result that works in half the population is a result with an unexplained variable in it. Nothing here has been tested in humans.
