Lab-grown blood cells for dogs edge toward a donor-free supply

Osaka researchers coaxed dog stem cells into hemoglobin-carrying blood cells, a first step past reliance on donor dogs.

Researchers at Osaka Metropolitan University, led by Professor Shingo Hatoya, report a way to generate red blood cell-like cells from canine induced pluripotent stem cells. The work appears in Stem Cells Translational Medicine, and the dog iPSCs were developed with TOKIWA-Bio Inc.

The team grew the stem cells as clusters to imitate how blood forms in the body, yielding progenitor cells and cells carrying hemoglobin. To follow the process, they used CRISPR-Cas9 to tag the red-cell marker glycophorin A so cells glow green when it appears, giving a real-time read on differentiation. Under tuned conditions, more than 96% of the analyzed cells expressed the marker — but only about 3% shed their nucleus, the enucleation step that defines a mature mammalian red cell.

The need here is a stubborn one. Veterinary blood banks barely exist, so transfusions for dogs lean on donor animals and are complicated by canine blood types. A route to manufacturing blood cells from stem cells could ease that scarcity over time, and the researchers say the same platform may inform human iPSC-derived blood products.

This is a starting line, not a finished therapy. The cells are not yet mature or fit for transfusion, only a small fraction enucleate, and the authors flag more work to raise the yield of functional cells and to understand differences between cell lines. What to watch is whether that 3% figure climbs in follow-up studies.
