Semaglutide extends female mouse lifespan by 92 days in aging study

Semaglutide treatment increased median lifespan in female mice by 12.4% in aging study

A UC Berkeley study published in *Nature* found semaglutide injections extended median lifespan in non-obese, non-diabetic female C57BL/6 mice by 92 days (12.4%), raising it from 742 to 834 days. The treatment began at 20 months of age, with 40 mice receiving daily semaglutide injections versus 39 saline controls. Semaglutide reduced food intake by 24% and shifted body composition toward lower fat and higher lean mass. It also improved motor coordination, glucose tolerance, and neurogenesis in the dentate gyrus while reducing inflammatory markers and senescence markers in key tissues. Mitochondrial gene expression increased in muscle tissue, and hematopoietic stem cells showed partial reversal of age-related myeloid bias.

The findings suggest semaglutide may counteract multiple aging pathways—though mouse lifespan (742 days) represents the lower end of typical female C57BL/6 mouse longevity. Human clinical trials for GLP-1 receptor agonists remain inconclusive, and effects may not translate to longer-lived mouse cohorts. Semaglutide does not demonstrate direct senolytic activity, as reduced senescence markers do not confirm cellular senolysis. This research provides mechanistic insights for future human therapies but does not yet indicate immediate health benefits for people.
